Depends on the character. Some characters for Stephanie 5 require that you actually own Stephanie 5 to use the skin (in other words, they use S5 UV maps). You shouldn't need to own V5 (unless the character uses Victoria 5 UV maps, and I'm not aware of any Stephanie 5 characters that do).

Wow you know what , this whole time I thought that in order to get Stephanie5 or Aiko 5 or anything like that you had to get Victoria5 first and then branch off from there..but all you need is genesis. Thanks alot for clearing that up for me.

No problem...and you're not too far off, as that was the case for Generation 4 characters...both Aiko 4 and Stephanie 4 require Victoria 4.2.
But with Genesis, Genesis itself is the base figure, so the "iconic" characters like Aiko 5, Freak 5, David 5, etc. don't require any other purchase. The same is basically true with Genesis 2: the base figures are Genesis 2 Female and Genesis 2 Male, so you don't need to own V6 or M6 to use characters like Stephanie 6, Darius 6 etc.
